1. Add 39.50 gm powder to 1.0 liter distilled/purified water.
2. Gently heat and bring to boiling.
3. Autoclave at 15 psi pressure at 121°C for 15 minutes.
4. Cool to 45-50°C.
5. Add aseptically 5-10% sterile defibrinated blood and mix thoroughly.
6. Dispense into sterile petri plates.
