1. Add 9.00 gm powder to 1.0 liter of distilled/purified water and mix thoroughly.
2. Gently heat and bring to boiling.
3. Distribute the medium into 4 equal parts into test tubes.
4. In the first test tube no amino acid is to be added.
5. To the remaining 3 parts, add separately 3 L-amino acids, L-Lysine hydrochloride, L-Arginine hydrochloride and L-Ornithine hydrochloride to a final concentration of 0.5%.
6. Dispense in 3 -4 ml quantities in screw capped tubes.
7. Autoclave at 15 psi pressure at 121°C for 15 minutes.
Note : To avoid false alkalinization at the surface of medium ;add liquid paraffin to a height of about 5mm
before sterilization.
