Inoculate (isolated colony) the tubes by stabbing the butt to the center through 2/3 rds of the medium with the following and incubate at 37°C for 18 -24 hours
After 24 hours of incubation observe the tubes for growth, motility, hydrogen sulphide (H2S) production and indole production by addition of 3 - 4 drops of Kovacs reagent

(+) Motiliy : is observed as diffused growth outward from the stab line or turbidity of the medium.
(+) H2S : is observed by blackening of the medium or along the stab line.
(+) Indole : is indiacted by a red colour after addition of Kovacs Reagent.
