After addition of desired antibiotic solutions incubate the plates at 35 ± 2°C for 18-24 hours.(Cylinder plate assay method)
Observe the plates for zone of inhibition around the punched holes. Compare the zone sizes with the zone sizes of the reference antibiotic.

Directions
1. Add 25.50 gm powder to distilled/purified water.
2. Bring volume to 1.0 liter and mix thoroughly.
3. Gently heat and bring to boiling.
4. Autoclave at 15 psi pressure at 121°C for 15 minutes.
5. Dispense into sterile petri plates.
