Cultural Response
Inoculate and incubate the tubes at 37 ± 2°C for 18-24 hrs. Cultural response observed when subcultured on Soyabean Casein Digest Agar (SCDA) , after incubation at 42 - 44°C for 18-24 hrs.

Directions
1. Add 82.05 gm powder to 1000 ml distilled/purified water and mix thoroughly.
2. Gently heat and bring to boiling.
3. Do not autoclave.
4. Cool to 45-500C.
5. Aseptically, add 19.0 ml of Iodine solution (in 20gm iodine add 25gm potassium iodide in 100ml sterile distilled water.) and 9.5ml of 0.1% Brilliant Green solution.
6. If desired ,add Novabiocin supplement (18352).
7. Mix well to disperse precipitate uniformly.
8. Dispense into sterile test tubes.
