SRL`s FITC conjugation teaching kit is a simple and reproducible tool which teaches the student a quick and easy method of labeling a protein/antibody. Fluroscein isothiocyanate (FITC) conjugates of antibodies, lectins, hormones, and growth factors have been used in a variety of immunohistochemical and flow cytometry applications. The protocols have been optimized using a couple of proteins, but may be adapted to other proteins or antibodies
Fluorescein isothiocyanate (FITC) is an amine-reactive derivatives of fluorescein dye that have wide-ranging application as antibody and other probe labels for use in fluorescence microscopy, flow cytometry and immunofluorescence-based assays such as Western blotting and ELISA. FITC is one of the most common chemical used for labeling proteins and antibodies. It is reactive towards primary amine groups on proteins, peptides and other bio-molecules. Freshly prepared carbonate - bicarbonate buffer is ideal for setting up reactions. Reactions are most efficient at pH 9.0. Conjugation of the dye with the protein or antibody occurs within an hour and the excess dye can be removed by gel filtration or dialysis in the same conjugation buffer.
