Composition :
Tryptone : 2.00 gm/lit.
NaCl : 5.00 gm/lit.
K2HPO4 : 0.30 gm/lit.
Bromothymol Blue :0.08 gm/lit.
Agar :2.00 gm/lit.
pH at 25 °C : 6.8 ± 0.2
Storage : Store between 8-25 °C

Directions :
1.Add 9.38 gm to distilled/deionized water.
2.Bring volume to 1.0 liter and mix thoroughly.
3.Heat gently and bring to boiling.
4.Autoclave at 15 psi pressure at 121°C for 15 minutes.
5.Add 1% carbohydrate before or after sterilization depending on heat lability.
Applications:
For use with added carbohydrate for differentiation of Gram-negative microorganisms based on oxidation - fermentation patterns. |
